I. Background | |
HER2 基因?qū)儆谌吮砥どL因子受體家族之一(ErbB 家族),該家族由 EGFR、HER2、 HER3 和 HER4 四個(gè)高度同源的蛋白成員組成,控制著細(xì)胞生長、存活分化和遷移。與其 他三個(gè)成員不同的是,HER2 蛋白在本質(zhì)上是一個(gè)孤兒受體,缺乏特異性配體,是唯一 一 個(gè)可以與家族其他成員結(jié)合形成二聚體結(jié)構(gòu)的受體。ErbB2 是一種無配體受體,而 ErbB3 缺乏酪氨酸激酶活性。因此,ErbB2 和 ErbB3 僅在 ErbB 異二聚體環(huán)境中才有活性,而由 Nrg(神經(jīng)調(diào)節(jié)蛋白)配體驅(qū)動(dòng)的 ErbB2-ErbB3 異二聚體是細(xì)胞生長和轉(zhuǎn)化方面最普遍和 最有效的復(fù)合物。配體激活的 ErbB2-ErbB3 異二聚體信號傳導(dǎo)效力的基礎(chǔ)在于,這種二聚 體能夠非常有效地發(fā)出信號,既可以通過 Ras-ERK(細(xì)胞外信號調(diào)節(jié)激酶)途徑進(jìn)行增殖, 也可以通過 PI3K-Akt/PKB(磷脂酰肌醇-3 激酶-Akt/蛋白激酶-B)途徑進(jìn)行存活。此外, 這種受體二聚體可以逃避下調(diào)機(jī)制,從而延長信號傳導(dǎo)時(shí)間。 | |
