| I. Background | |
| CRF1R 屬于 B 類 GPCR 家族，在中樞神經(jīng)系統(tǒng)（下丘腦、杏仁核、前額葉皮層）及外 周組織（胃腸、皮膚）表達，是機體應(yīng)激應(yīng)答的核心調(diào)控者。CRF1R 的內(nèi)源性配體促腎上 腺皮質(zhì)激素釋放因子（CRF）和 尿皮質(zhì)素 1（UCN1）結(jié)合后，觸發(fā)受體構(gòu)象變化，優(yōu)先偶 聯(lián) Gs 蛋白，激活腺苷酸環(huán)化酶（AC），升高 cAMP，啟動 PKA/CREB 信號軸，驅(qū)動下丘 腦-垂體-腎上腺軸（HPA 軸）激活，最終促進 糖皮質(zhì)激素（皮質(zhì)醇）釋放以應(yīng)對生理/心理 壓力；同時通過β-arrestin 通路調(diào)節(jié)受體脫敏內(nèi)化。該受體過度激活與焦慮癥、抑郁癥、腸 易激綜合征（IBS）及藥物成癮密切相關(guān)。 | |
